The tumor microenvironment refers to the local complex environment (with characteristics such as hypoxia, glycolysis, low pH, relative lack of nutrients, and interstitial high pressure) in which tumors are located during the growth process, mainly including tumor cells, endothelial cells, immune cells, fibroblasts, pericytes and extracellular matrix, as well as soluble substances such as cytokines and peptide growth factors secreted by these cells. Tumor angiogenesis refers to the process in which the tumor microenvironment induces the formation of a capillary-based vascular system based on the original blood vessels, and establishes blood circulation in the tumor tissue.

When the tumor grows to 1-2 mm in diameter, it needs to establish an external tissue environment suitable for its own growth. At this time, cells in the microenvironment are in contact with each other, cytokines interact, and delicate and complex communication occurs between components. By inducing angiogenesis and other pathways, new nutrient metabolism networks are continuously constructed, thereby promoting further tumor growth.

To this end, we have started to establish a three-dimensional culture model (3D culture model) and successfully induced the formation of blood vessels from endothelial cells. Advances in the technique of culturing vascular endothelial cells in vitro and experimental methods of angiogenesis have promoted the research progress of angiogenesis. Since then, anti-angiogenic therapy has been aided in various aspects, such as attacking existing tumor blood vessels; neutralizing angiogenesis-promoting factors, thereby inhibiting angiogenesis; using endogenous or exogenous angiogenesis-inhibiting factors to inhibit angiogenesis.

There are a variety of imaging techniques that can be used to evaluate tumor angiogenesis. Each method has its own unique advantages and application scope, but all of them can non-invasively, rapidly, and evaluate tumor angiogenesis and anti-angiogenic efficacy in vivo. Through these technologies, more and better tumor vascular marker molecules are discovered, suitable targets are found to block and destroy tumor angiogenesis, and effective anti-tumor angiogenesis drugs are developed.
