Introduction

Immune Signaling in TME

With the widespread application of molecular biology techniques in tumor immunology research, the study of tumor immune response, immune tolerance and immune regulation mechanism has gradually risen to the molecular level, and the study of antigen receptors and other cell membrane molecule-mediated signal transduction processes has become the main frontier of contemporary immunology research. At present, a variety of signal transduction pathways have been found to be related to the immune regulation of tumors, and the most studied ones are TLRs signaling pathway, JAK-STAT signaling pathway, T-cell activation pathway, B-cell activation pathway and so on. However, the research on immune signaling pathways in tumor microenvironment (TME) is still limited, and the specific signal transduction mechanism of single pathway is not fully clarified, and the interconnection between the pathways is even less studied. Therefore, many immune mechanisms of tumors are still hypothesized and need further research to be confirmed.

Solutions

Since tumor immune response, immune tolerance and immune regulation are involved in tumorigenesis, development and regression, it is imperative to investigate the pathways and molecular mechanisms of tumor immune signaling and to find potential therapeutic targets for tumor immunotherapy. With the increasing emphasis on precision tumor therapy, more and more targeted drugs, including angiogenesis inhibitors, immunomodulators and signal transduction inhibitors, are being used in clinical practice.

Alfa Cytology has established an innovative technology platform for tumor microenvironment centers and is developing multiple technologies to provide high quality solutions and services to our global collaborators. In-depth studies of intracellular signaling of immune signals in the tumor microenvironment (TME) or intrinsic immunity / adaptive immune response / immune effector molecule related signaling, we expect to identify new targets and develop new approaches for combination therapy and reduction of side effects.

Key technologies

Flow cytometric immunofluorescence (FCM) / immunofluorescence lmmunofluorescence (IF). Flow cytometric immunofluorescence (FCM) can be used to detect protein molecules in different cell subpopulations from the single cell level, and two different fluorescent dye-labeled antibodies against different protein molecules can be used to detect two different molecules simultaneously in the same cell ( Double lF), or multi-parameter The detection of multiple intracellular molecules can also be performed by multi-parameter flow cytometry.

The expression of the receptor is regulated by treatment of the receptor with antibodies or RNAi / overexpression, and then the expression of each gene of the downstream pathway is studied by gene microarray technology.

Fluorescence resonance energy transfer (FRET) is an effective method for qualitative and quantitative detection of interactions between biological macromolecules, and is a dynamic monitoring of the acceptor-donor interactions of tumor immunity in living cells in real time.
