Knockouts are manipulations of DNA that result in the loss of function or complete loss of the corresponding protein. Knockout models are "loss-of-function" mutants. Generally, they can be divided into three types: conventional knockout models, conditional knockout models and KO first (condition-ready) models.

Random transgenic models can be obtained by transposon-mediated insertion, DNA microinjection and lentivirus-mediated transfection methods. Exogenous genes can be targeted into embryonic or ES cells in the form of point mutations and cassette insertions (e.g., reporter genes), resulting in "gain-of-function" mutants.

Gene knockdown is a technique used to reduce the expression of one or more genes in an organism, resulting in temporary functional neutralization. This can be achieved through gene modification or the use of short RNA oligonucleotides (e.g. siRNA) that are complementary to the target mRNA to be silenced.

The ES cell-mediated targeting technique for obtaining gene editing mouse models is more suitable than the CRISPR/Cas9 technique for making mouse models with complex editing strategies or large insertion fragments.

CRISPR/Cas9 technology is the most commonly used gene editing technology for the preparation of CKO/KO/KI/PM mice with specific gene modifications.

Genetically engineered mouse models are obtained by random integration of DNA into the mouse genome and are generally used to obtain the preparation of overexpressing or conditionally overexpressing mice. With the increasing efficiency of K, most investigators have switched to the use of H11 or Rosa26 locus targeted knock-in for the production of overexpression mouse models.
