Solutions for NK immunomodulation in the TME

Relying on the technology platform of the tumor microenvironment center, Alfa Cytology provides global collaborators with solutions and related services to study the relationship between tumor immunity and natural killer cells (NK) in the tumor microenvironment (TME), helping them to rapidly achieve their research goals.

Introduction

Immunomodulation of NK in TME

NK cells (natural killer cells) belong to a special class of lymphocytes. Unlike T cells, they can recognize and kill their own mutated cells without prior sensitization, and are important cells for innate immunity. The role of NK cells in the development of malignant tumors is very important. This process of action is accomplished through the binding of specific molecules (ligands) on the surface of mutant cells by receptors on the surface of NK cells. The discovery of new immune molecules that regulate the killing function of NK cells on cancer cells and their regulatory mechanisms not only provide new perspectives and ideas for an in-depth understanding of the immune escape mechanism of tumor cells, but also provide an important theoretical and experimental basis for tumor immunotherapy research.

Solutions

During tumor cell development, NK (natural killer) cells, which are important components of the intrinsic immune system, not only kill tumor cells directly by secreting perforin and granzyme, but also regulate the function of other immune cells by secreting cytokines, thus playing an important role in anti-tumor immunity. On the other hand, while immune cells attack tumor cells, tumor cells also generate multiple mechanisms to escape from the surveillance of the immune system. Tumor cells can directly or indirectly regulate the function of immune cells through secreted soluble factors or ligands expressed on their surface to inhibit the killing of tumor cells by immune cells, thus maintaining the survival development of the tumor itself. Therefore, how to regulate the killing activity of NK cells on tumor cells and enhance the sensitivity of tumor cells to NK cell killing is a key issue in current research on tumor immunotherapy.

Alfa Cytology has established a tumor microenvironment technology platform and developed several emerging technologies through which it helps global collaborators to deeply explore the relationship between tumor immunity and NK (natural killer) cells in the tumor microenvironment (TME), hoping to find potential targets for tumor therapy in the process and provide new ideas for clinical anti-tumor treatment.

Key technologies

Microfluidic technology can achieve single-cell manipulation at the order of magnitude, and has become one of the most promising single-cell analysis techniques in recent years. The application of microfluidic technology in single-cell pairing, fusion, migration, and cell-to-cell immune response is discussed, and the application prospects of microfluidic microarrays are prospected with organ-on-a-chip as an example. Using this technology, it is possible to deeply study the relationship between tumor immunity and NK (natural killer) cells in the tumor microenvironment (TME), hoping to find potential targets for tumor therapy in the process and provide new ideas for clinical anti-tumor treatment.

Alfa Cytology offers flow cytometry, ELISA/ELISPOT, hoechst staining, immunofluorescence assays, western blot and other techniques that are classical methods for studying tumor immunity. Combined with these techniques, experiments are performed from various aspects / perspectives to elucidate the molecular mechanisms of functional changes of NK (natural killer) cells in the microenvironment and how they exert immune regulatory functions.
