Introduction

Immunomodulation of TAM in TME

Tumor-associated macrophages (TAM), macrophages that evolve from infiltration of peripheral blood mononuclear cells into solid tumor tissue, account for a large proportion of tumor stromal cells and are receiving increasing attention. It is a heterogeneous cell population that exhibits a variety of phenotypes depending on the type of tumor and its location in the tumor microenvironment (TME). It is mainly considered to have pro-tumorigenic effects. In addition to pro-angiogenic and metastatic effects, these include immunosuppressive effects. However, how TAM is specifically formed, how the phenotypic changes of TAM are regulated, what is the relationship between the biological behavior of tumor cells and TAM, and how TAM exerts its immunomodulatory effects. These questions are still not clearly answered. Therefore, elucidating the molecular mechanisms of functional changes of macrophages in the microenvironment will likely provide new clues for future tumor biotherapy.

Solutions

Previously, tumor-associated macrophages (TAM) were thought to be important anti-tumor effector cells that could either directly kill tumor cells or induce an immune response by presenting tumor-associated antigens to clear tumors. However, in recent years, more and more studies have shown that TAM do not exert anti-tumor effects directly, but act indirectly on tumor-associated stroma or endothelial cells by secreting specific cytokines and growth factors. Further, it is involved in processes such as pro-tumor development, stimulation of tumor cell growth and metastasis, induction of tumor neovascularization and lymphangiogenesis, and immunosuppression. These have suggested to researchers that TAM may be a new target for human intervention in the treatment of tumors. In-depth study of TAM has the potential to thoroughly elucidate the mechanisms of tumor immune tolerance and explore effective immunotherapeutic approaches.
