Introduction

The construction of a tumour microenvironment (TME) to study tumour heterogeneity, tumour progression and subsequent metastasis, drug response and drug resistance has been a great challenge in cancer research. There are two effective research models, patient-derived xenografts (PDX) and patients derived organoids. Patient-derived organoids are miniature 3D tumor cell models that are cultured in the laboratory from primary tumors in patients with the disease.

Patients derived organoids can encapsulate the tumor microenvironment including tumor cells and non-tumor host components, and are phenotypically and genetically similar to the tumor tissue from which they originate, highly mimicking the characteristics of the source tumor tissue, preserving tumor heterogeneity between individuals, and can be used for mechanistic studies and functional testing.

Service content / steps

1. 2D preculture

Organoids are derived from primary cells (i.e. intestinal, lung or kidney cells) or induced pluripotent stem cells.

2. Development into 3D organoids

The medium includes extracellular matrix (ECM) proteins and different growth factors, which will vary depending on the type of tissue being cultured.

3. Organoid culture

Organoid culture is a lengthy process that may involve different steps and the use of different media. During this process, the health of the cells needs to be monitored (imaged), generally to understand developmental biology and tissue.

4. Monitoring and readings

With high-intensity imaging, it is possible to monitor and observe the growth and differentiation of organoids, 3D reconstruction of structures, complex analysis of organoid structures, cell morphology and activity, and the expression of different cellular markers.

5. Confocal imaging and 3D analysis

Confocal imaging and 3D analysis of organoids allows the observation and quantification of organoids and the cells that make up organoids. Multiple quantitative descriptions of organoids can be used to study disease phenotypes and compound effects.
